'Calculate cost from units' means that the price of a resource assignment is calculated from one of the five available prices in the resource dictionary, rather than entered manually for each resource assignment. I ALWAYS tick this box, since it can be overridden for a particular resource assignment if there is a fixed price you must record independent of the resource quantity.
'Auto Compute Actuals' is ONLY appropriate if you do not have a timesheet system to record the real quantity of Actual aka expended quantities for each resource assignment. This fixes for the entire project a CPI of 1.00.
