I presume you are using MS Project 2003 Server and want to upload a project that was created under a valid MS Project Server connection (i.e. not a "local project")
1. Click File, Save; by default this project will be saved in the MS SQL Server database, or
2. Click Collaborate, Publish ... MS Project will advise that the project should be saved first and request you to agree.
